Abstract: Three novel anti-diabetic drugs, namely, 2-hydroxy-1-naphthaldehyde (HNPD), 3,6-diamino acridine hydrochloride (AHCL), and 4-oxo-4H-benzo[h]chromene-3- carbaldehyde (BCC), were evaluated to understand their inhibition properties against the diabetic enzymes (alpha-glucosidase and alpha-amylase) using a unique integrated in silico and in vitro workflow. These drug candidates were developed through the combination of two-dimensional techniques, namely, in silico molecular docking and in vitro spectrofluorometry. The in silico molecular docking and in vitro spectrofluorometry experiments established quantitative and qualitative interactions, respectively, between the compounds and the target proteins. In silico experiments showed that AHCL (6.1 Kcal/mol) and BCC (5.6 Kcal/mol) exhibit the highest binding affinities to two important proteins associated with diabetes. All in silico and in vitro data were validated and compared with a similar experimental dataset with the control drug Acarbose. The compound 4-oxo-4H-benzo[h]chromene-3-carbaldehyde (BCC) was synthesised in the laboratory and characterised by MIR analysis. This test workflow uses a special combination of techniques to standardise the intrinsic fluorescence properties of drug candidates and target proteins, replacing methods with significant computing costs and a strong validation strategy, namely, alpha-glucosidase (PDB id 7KBJ) and alpha-amylase (PDB id 5U3A). Further, our unique methodology could identify new novel anti-diabetic drug candidates with minimum turnaround time.
