Abstract: An automated imaging and analysis system for microbiological culture plates comprising a multi-camera stereo imaging assembly, multi-spectral LED ring illumination, and an AIpowered computer vision processing pipeline. The system includes a perpendicular 12- megapixel top camera and two oblique 8-megapixel cameras positioned at 45-degree angles with a 150mm stereo baseline, enabling three-dimensional reconstruction of bacterial colonies with depth resolution of +0.1mm. A 40-element LED ring provides selectable illumination across white, ultraviolet, red, green, and blue wavelengths with individual PWM intensity control. A novel 360-degree radial zone profiling algorithm casts 360 intensity rays from each antibiotic disk center to precisely measure zones of inhibition, detect heteroresistance, and classify isolates as susceptible, intermediate, or resistant. The software pipeline performs image preprocessing, contour-based segmentation, 2D morphological feature extraction across 67 configurable parameters, and optional 3D reconstruction via stereo vision or photometric stereo methods. Quality control validation assesses lawn uniformity, disk spacing, and contamination. Results are stored in a SQLite database and exported in CSV, PDF, or LIMS-compatible formats within approximately 3.2 seconds per plate. The system is housed in a light-controlled enclosure with an integrated 7-inch touchscreen interface.
