Abstract: A microbiome-responsive drug delivery system (100) for localized and on-demand delivery of active agents at sites of microbial dysbiosis. The system (100) comprises a nanoparticle core matrix (101) having a particle size between 50 nm and 500 nm, an active payload (102) encapsulated within the nanoparticle core matrix (101), a responsive outer coating (103), a sensor element (104) configured to selectively recognize quorum-sensing molecules secreted by pathogenic microorganisms, a cleavable linker (105), and a microbiota-preservation mechanism. Upon recognition of the quorum-sensing molecules, the sensor element (104) activates cleavage or structural alteration of the cleavable linker (105), thereby disrupting the responsive outer coating and releasing the active payload in a localized manner. The system may further comprise a biofilm-responsive component and skin-barrier restoration agents. The invention provides targeted drug release, preservation of beneficial skin microbiota, improved bioavailability, enhanced therapeutic efficacy, and reduced adverse effects for treatment of microbial dysbiosis-associated skin disorders.
