Abstract: ABSTRACT Disclosed herein is a Mycoplasma Agglutination Test (MAT), a novel, rapid, and field-applicable serological diagnostic method for the detection of Contagious Caprine Pleuropneumonia (CCPP) in goats. The method employs whole-cell Mycoplasma capricolum subsp. capripneumoniae antigen stained with tetrazolium (blue-black) or neotetrazolium (red-pink) dye to produce a visually distinct agglutination reaction upon mixing with antibody-positive caprine serum. The test requires only 20 µL of serum and delivers results within 3 minutes, with no need for electricity, optical instruments, or specialized laboratory infrastructure. Validation against competitive ELISA (cELISA) demonstrated a sensitivity of 80.65% and specificity of 75.36%, confirming suitability for first- line screening in resource-limited and endemic settings. The MAT represents a significant advancement in accessible livestock disease diagnostics, enabling rapid, on-site detection and facilitating timely implementation of disease control measures.
