The application for the patent was published on August 28, 2026, under issue no. 35/2026.
Abstract: ABSTRACT MULTI-PLATFORM BACTERIOPHAGE LYSIN COMPOSITIONS FOR ANTIBIOTIC-RESISTANT INFECTIONS AND DISSOLVING BIOFILMS The disclosure provides a multi-platform anti-biofilm system for antibiotic-resistant infections, comprising twenty-three nucleotide constructs (SEQ ID NOs: 1–23) across three delivery architectures. The first (SEQ ID NOs: 1–5) comprises CAR-macrophage lentiviral and mammalian expression vectors encoding Anti-ClfA4 scFv chimeric receptors fused via a CD8 hinge and PD-1 transmembrane domain to secreted lytic payloads, including cpl-1 amidase, cpl-7 endolysin, SAL-1 CHAP domain, SMAP-29, LysPA26, and Dep37 depolymerase. The second (SEQ ID NOs: 6–13 and 18–23) comprises pT181 rolling-circle plasmids and EcoRI/KpnI-flanked cassettes encoding surface-anchored or secreted lysins for delivery from commensal Staphylococcus epidermidis and Lactobacillus plantarum chassis, with interchangeable signal peptides. The third (SEQ ID NOs: 14– 17) comprises hEF1-HTLV-driven exosomal constructs routing lytic cargo into HEK293-derived vesicles via a DLG4 ExoTag, or displaying endolysins via a C1C2 phosphatidylserine-binding domain. Manufacturing methods and compositions are also provided.
