Abstract: ABSTRACT MULTI-MODULE ENGINEERED EXTRACELLULAR VESICLE COMPOSITIONS AND CLEAN EXOSOME FACTORY SYSTEMS FOR HAIR GROWTH AND HAIR FOLLICLE REGENERATION The instant disclosure provides engineered extracellular vesicle compositions for hair follicle regeneration comprising four cooperatively acting therapeutic modules delivered within exosomes produced by genetically modified producer cells. A growth factor module comprising FGF7, FGF10, and FGF2 stimulates epithelial and dermal papilla cell activity. A receptor amplification module comprising FGFR2IIIb augments hair follicle cell responsiveness to fibroblast growth factor signalling. A stem cell activation module comprising Wnt3a and Lhx2 regulates follicular stem cell identity and maintenance. An immune privilege restoration module comprising PD-L1, IL-10, IL-4, and Annexin A1 modulates the perifollicular immune microenvironment. The disclosure further provides lentiviral expression constructs with a tetracycline-responsive bleomycin resistance cassette activated by a non-integrated transactivator during selection and rendered permanently silent upon transactivator degradation, enabling stable clean exosome factory producer cell lines, methods of producing enriched extracellular vesicles, and formulations for delivery by intradermal injection, topical application, or microneedle-assisted transdermal delivery.
