Abstract: The present invention discloses a novel fixed-dose combination (FDC) multi-layer tablet comprising Clopidogrel (50 mg), Apixaban (5 mg), and Rosuvastatin (10 mg). This multi-layer tablet features an immediate-release top layer of Clopidogrel, an extended-release core of Apixaban, and an intermediate layer of Rosuvastatin encapsulated in a protective coating. The formulation ensures tailored drug release profiles: rapid dissolution of Clopidogrel within 6.5–8 minutes, intermediate release of Rosuvastatin over 6 hours, and sustained release of Apixaban over 19–20 hours. By addressing formulation challenges, such as the acid-labile nature of Rosuvastatin and potential drug interactions, this invention offers a stable, effective, and convenient solution for the prophylactic management of atherosclerosis and coronary thrombosis.
