Abstract: ABSTRACT MS-BASED ASSAYS TO QUANTIFY A PANEL OF TARGETED SERUM PROTEIN BIOMARKERS FOR DETECTING COLORECTAL CANCER The present disclosure relates to a mass spectrometry-based method for identification and 5 quantification of colorectal cancer-specific protein biomarkers; a composition of proteotypic peptides for identification of colorectal cancer-specific protein biomarkers; and a mass spectrometry assay-based system for identification and quantification of colorectal cancer-specific protein biomarkers. More particularly, the mass spectrometry-based method comprises steps of: identification of potential protein biomarkers by 10 literature search; detection of differentially expressed proteins and neoantigens for colorectal cancer by data-independent acquisition (DIA) analysis; selection of proteotypic peptides for literature-curated protein biomarkers , differentially expressed proteins and neoantigens from DIA analysis; and optimization of a parallel reaction monitoring (PRM) assay with Natural (NAT) and Stable-Isotope- Standard (SIS) peptides proteotypic 15 peptides. The disclosed method enables a minimally- invasive approach to CRC detection and differentiation of CRC from Inflammatory Bowel Diseases (IBD) with a panel of 15 protein biomarkers.
