Abstract: The invention relates to a moisture-activated patch for local-anesthetic delivery. The patch comprises a backing layer, a compressed tablet core, and a tissue-facing release layer. The compressed tablet core contains lidocaine, benzocaine, prilocaine, tetracaine, or a combination thereof and a water-swellable matrix former selected from HPMC, NaCMC, PVA, carbopol, or chitosan. Water, saliva, wound exudate, or tissue fluid reaches the compressed tablet core and swells the matrix former, thereby initiating controlled release through the tissue-facing release layer. Release is sustained for a period ranging from 15 minutes to 8 hours. A penetration- enhancing agent may be included for transdermal or transmucosal delivery. The patch is suitable for application to oral mucosa, skin, or wounds and is applicable to pharmaceutical manufacture and local pain-management products.
