Abstract: The present disclosure provides a modified polymer conjugate for targeted cancer delivery comprising folic acid conjugated with phenylalanine through an amide bond to form a phenylalanine-modified folic acid, and methoxy polyethylene glycol conjugated to the phenylalanine-modified folic acid through a succinic acid linker. The modified polymer conjugate is configured for targeting folate receptors overexpressed on cancer cells. The phenylalanine conjugation enhances binding affinity of folic acid to folate receptors, and the methoxy polyethylene glycol modification improves solubility, stability, and pharmacokinetic performance. A method of synthesizing the modified polymer conjugate is also provided, comprising conjugating folic acid with phenylalanine to form a phenylalanine-modified folic acid, activating methoxy polyethylene glycol with succinic anhydride to form an mPEG-acid, and conjugating the phenylalanine-modified folic acid with the mPEG-acid.
