Abstract: ABSTRACT The present invention relates to an MMP-8-responsive therapeutic mucoadhesive hydrogel for site-specific management of oral inflammation. The hydrogel comprised a biocompatible hydrogel-forming polymer, a mucoadhesive polymer, an MMP-8-responsive proteolytically cleavable component, a therapeutic active agent and a pharmaceutically acceptable aqueous vehicle. The formulation was developed to overcome rapid removal of conventional oral topical formulations by saliva and mechanical movement and to provide prolonged residence at inflamed oral tissues. The MMP-8-responsive component was incorporated to provide enzyme-triggered therapeutic release in an inflammatory microenvironment exhibiting elevated MMP-8 activity. The optimized hydrogel demonstrated suitable physicochemical characteristics, including homogeneous appearance, acceptable pH, viscosity, spreadability, swelling behavior, drug-content uniformity and mucoadhesive strength. In-vitro evaluation demonstrated sustained drug release under physiological conditions and enhanced release in the presence of MMP-8. Biological evaluation demonstrated improved anti-inflammatory activity compared with the free therapeutic agent and conventional non-responsive hydrogel. The invention therefore provides a localized, prolonged and inflammation-responsive platform for management of oral inflammatory and periodontal disorders.
