Abstract: A mitochondria-targeted nano co-amorphous composition is disclosed, comprising curcumin and mitoquinone mesylate at a molar ratio of 2:1 to 1:2, wherein the said curcumin and mitoquinone mesylate constitute a single-phase amorphous molecular dispersion stabilised by multi-point intermolecular hydrogen bonding involving the enolic hydroxyl of curcumin and the methoxy, quinone carbonyl and methanesulfonate sulfonyl oxygens of mitoquinone mesylate. The composition exhibits a single glass transition temperature in the range of 95 to 130 degrees Celsius, broad amorphous halos in powder X-ray diffraction, at least five-fold enhanced equilibrium aqueous solubility of curcumin, suppression of recrystallisation under ICH Q1A(R2) long-term and accelerated stress, multi-fold enhancement of oral bioavailability in rat pharmacokinetic studies, mitochondrial accumulation of co-amorphised curcumin, and a favourable therapeutic window.
