Abstract: The present invention discloses a catalyst-free and microwave-assisted amination process for the synthesis of biologically active heterocyclic compounds. The process involves reacting an electron-rich alkoxy-substituted chloro-pyrimidine with a secondary amine in the presence of N,N-diisopropylethylamine (DIPEA) and PEG-400 solvent. The reaction mixture (100, 200, 300) is subjected to microwave irradiation at 110 °C for a compressed duration of 8 to 30 minutes (101, 201, 301). The process entirely eliminates the requirement for transition-metal catalysts, specialized ligands, and inert atmospheric conditions. Following reaction completion and cooling (102, 202, 302), the product is extracted and purified via recrystallization in isopropyl alcohol (103-106, 203-206, 303-306) to afford highly purified aminated compounds. The methodology offers a sustainable, cost-effective, and rapid synthetic platform for pharmaceutical synthesis.
