Abstract: ABSTRACT [0017] The present invention relates to a point-of-care diagnostic device for the direct detection of bacterial antimicrobial resistance (AMR) genes from a patient sample, without requiring bacterial culture or nucleic acid sequencing. The device comprises three integrated elements: (i) a microneedle patch that minimally- invasively samples a microliter-scale volume of interstitial fluid or capillary blood; (ii) a disposable electrode chip pre-loaded with a multiplexed CRISPR-based detection panel (Cas12a and/or Cas13 effectors, each paired with a guide RNA specific to a target AMR gene such as mecA, blaNDM, or vanA), so that a single sample is screened against several resistance genes simultaneously; and (iii) a gene-specific electrochemical reporter system, in which each detection channel is tagged with a distinct redox mediator, allowing multiple resistance-gene signals to be distinguished and read out on one chip using a simple potentiostat (including smartphone- dongle-scale readers). Collectively, this integration collapses sample collection, target amplification/detection, and multiplexed readout into a single point-of-care workflow, in contrast to existing CRISPR diagnostic platforms (e.g., SHERLOCK, DETECTR) which typically require separate venous sample collection, fluorescence or lateral-flow readout limited to single or few targets, and often laboratory-based reader equipment
