Abstract: The present invention relates to a microfluidic cartridge for sequential enzymatic detection of multiple edible oil adulterants using passive capillary flow for rapid and on-site authentication of edible plant oils. An edible oil sample is introduced into the cartridge through a sample inlet and is transported through interconnected microchannels using passive capillary forces without the need for external pumps or power sources. The cartridge contains pre-stored enzymatic reagents that are automatically reconstituted upon contact with the flowing sample. Passive flow control structures regulate the sequential movement of the sample through multiple reaction chambers, enabling controlled enzymatic reactions in a predetermined order. The reaction products are directed to an integrated detection chamber, where analytical signals corresponding to the presence of edible oil adulterants are generated. The cartridge is capable of multiplex detection of multiple adulterants from a single sample while minimizing reagent consumption and sample handling. The integrated lab-on-a-chip platform combines sample introduction, passive fluid transport, reagent storage, sequential enzymatic reactions, and detection within a compact disposable device. The passive capillary flow architecture ensures reliable and autonomous fluid movement while reducing operational complexity. The integrated reagent storage eliminates manual reagent preparation and improves the consistency of enzymatic analysis. The sequential arrangement of reaction chambers minimizes analytical interference and enhances detection specificity for multiple adulterants. The compact disposable design enables rapid analysis with minimal sample volume and reduced operational cost. The invention provides a portable, user-friendly, and cost-effective solution for edible oil authentication, reducing dependence on sophisticated laboratory equipment and enabling reliable food quality monitoring for food safety, quality assurance, and field-based applications.
