Abstract: The present invention discloses a process for recovery, enrichment, and microencapsulation of spermidine from cooking liquor generated during blanching of Calocybe indica (milky white mushroom). The process comprises membrane filtration, low-temperature vacuum concentration, and food-grade enrichment to obtain a spermidine-rich fraction with an enrichment factor of up to 7.5-fold and overall recovery of 72%. The enriched spermidine is encapsulated using a soy protein isolate– dextran and sodium alginate external gelation water-in-oil-in-water double emulsion system, achieving encapsulation efficiencies of 92–99%. Subsequent freeze-drying produces stable microcapsules with water activity below 0.23, a porous network microstructure, and retained encapsulation efficiencies of 56–98%. The invention simultaneously facilitates mushroom de-bittering and valorization of an otherwise discarded processing by-product, converting the cooking liquor into a functional ingredient rich in spermidine. The resulting microcapsules are suitable for incorporation into ready-to-eat foods, packaged cooking-liquor formulations, dietary supplements, and nutraceutical products.
