Abstract: The present invention relates to a computer-implemented method for identifying and validating repurposable therapeutic candidates common to Alzheimer's disease and Huntington's disease. Multiple independent transcriptomic datasets are subjected to cross-disease meta-analysis to identify commonly dysregulated genes, which are analysed through protein-protein interaction network construction, hub-gene topology analysis, and functional enrichment to identify a common molecular target. Disease-associated transcriptional signatures are queried against a perturbational transcriptomic database to identify compounds predicted to reverse the disease- associated expression profile and are further filtered using clinical trial information. Shortlisted compounds are docked into a predicted binding site of the common target, and a selected target-ligand complex is evaluated using all-atom molecular dynamics simulation. Structural stability, interaction persistence, binding free energy, and per-residue energy contributions are determined to validate the candidate. The method provides an integrated cross-disease computational framework for therapeutic target identification and drug repurposing.
