Abstract: The present invention relates to plant-mediated green synthesis of copper oxide (CuO) nanoparticles using a methanolic extract of Juglans regia L. roots. Cupric nitrate trihydrate is used as a copper precursor and Tween 80 as a surfactant component. The root extract is introduced into the heated precursor mixture, followed by controlled stirring, solvent evaporation, calcination, and grinding to obtain monoclinic CuO nanoparticles. The resulting nanoparticles exhibit a representative mean crystallite size of approximately 16.22 nm and surface-associated plant-derived capping characteristics. The CuO nanoparticles demonstrate visible-light photocatalytic degradation of methylene blue, antibacterial activity against representative Gram-positive and Gram-negative bacterial strains, and a differential in vitro cytotoxic response between SH-SY5Y and 3T3-L1 cells. The disclosed synthesis thereby provides multifunctional plant-mediated CuO nanoparticles for environmental- remediation, antibacterial, and further biomedical investigations.
