Abstract: Acute myeloid leukemia (AML) is an aggressive blood cancer for which the MV4-11 cell line is a widely used laboratory model for evaluating candidate drugs, typically through measurement of the half-maximal inhibitory concentration (IC50) in cell-viability assays. This invention provides a computer-implemented method for screening anti-leukemic drug candidates against MV4-1 1 cells using spectral graph-theoretical molecular descriptors. Each compound is represented as a molecular graph, and the Estrada index is computed from the eigenvalues of its adjacency matrix as the principal descriptor of molecular structure, optionally supplemented by distance spectral energy and topological indices including the Wiener, hyper-Wiener, Harary, and eccentricity-based indices. These descriptors are correlated with experimentally determined MV4-11 IC50 values through a QSPR model, enabling the IC50 of untested compounds to be predicted and ranked prior to laboratory assay. The method reduces the number of compounds requiring costly cell-based screening, accelerates the identification of promising antileukemic leads, and may be retrained on the cytotoxicity data of other cell lines to extend its use to broader cancer drug discovery.
