Abstract: ABSTRACT Method for rapid isolation of DNA from plant tissues The present invention discloses an optimized universal method for extracting high-quality genomic DNA from diverse plants types and various parts of plant with minimal sample preparation, lesser cost per sample and optimized time of 60 minutes. The three staged method includes a lysis stage for rupturing and disintegrating the cell wall to release their intracellular organelles including genomic DNA and proteins, a precipitation stage for precipitating the plant DNA from lysed tissue samples, and a washing and elution stage for final elution of the DNA pellet. The invention uses a novel lysis buffer containing CTAB, Triton X, Sodium Chloride, Sodium Metabisulfite, 2-Mercaptoethanol, and Proteinase K, a unique combination that enhances the breakdown of cell walls, denatures proteins, neutralizes phenolic compounds, and dissolves polysaccharides. The method allows for reliable and reproducible DNA extraction, facilitating genomic studies and forensic applications. Fig. 1
