Abstract: The present invention discloses a method (100) for preparing a hyaluronic acid mediated chitosan-coated Mogroside-V-Melphalan nanoconjugate for CD44-targeted ovarian cancer therapy. The method comprises preparing an organic phase comprising 10 mg of a Mogroside-V-Melphalan conjugate and 20 mg of native Mogroside-V dissolved in 2 mL of 70% (v/v) ethanol, followed by precision-controlled nanoprecipitation into 5 mL of an aqueous phase under ultrasonication to form a stabilized anionic Mogroside-V nanocore at pH 3. The nanocore is sequentially coated with 7 mL of 0.1% (w/v) chitosan at pH 4.5 and 14 mL of 0.05% (w/v) hyaluronic acid at pH 5.5 through electrostatic self-assembly to form a multilayered nanoconjugate. A final stabilization protocol comprising 10 cycles of ultrasonication produces a water-dispersible nanoconjugate, wherein the hyaluronic acid outer layer facilitates CD44 receptor-mediated targeting, the chitosan layer provides electrostatic stabilization, and the self-assembled Mogroside-V nanocore encapsulates the Mogroside-V-Melphalan conjugate for controlled release.
