Abstract: ABSTRACT The present invention provides an efficient and general method for the reductive etherification of carbonyl compounds with alcohols using chlorodimethylsilane (CDMS) in the presence of a commercially available thiourea organocatalyst. This method enables the selective formation of ether bonds across a wide range of alcohols, including primary, secondary, and tertiary alcohols, while tolerating various functional groups such as esters, halides, olefins, alkynes, nitro, cyano, trimethylsilyl, amides, and carbamates. Utilizing chloride anion binding catalysis, the thiourea catalyst enhances oxocarbenium ion electrophilicity, facilitating efficient hydride transfer from CDMS. The method is applicable to the late-stage functionalization of complex molecules, including natural products like steroids, terpenoids, peptides, and carbohydrates, enabling drug modification. Notably, the hydroxyl group in Ritonavir was selectively alkylated to generate novel derivatives. This robust and scalable approach provides a valuable tool for medicinal and synthetic chemistry applications.
