Abstract: The present invention discloses a method (100) for preparing a Mogroside-V–Melphalan ester conjugate, comprising steps of mechanically triturating 22 mg of Mogroside-V with 50 µL of glycerol to obtain a glycerol-plasticized Mogroside-V mixture; adding 50 µL of 1 M citric acid to the glycerol-plasticized Mogroside-V mixture; adding 30 mg of Melphalan to the mixture; mechanically grinding the resulting mixture using a pestle and mortar for 15-20 minutes to obtain a substantially homogeneous viscous reaction paste; thermally curing the viscous reaction paste at 55°C for 1 hour, thereby promoting esterification between at least one hydroxyl group of a glycosidic moiety of Mogroside-V and a carboxyl group of Melphalan to form the Mogroside-V-Melphalan ester conjugate; and lyophilizing the thermally cured reaction paste overnight to remove residual moisture and obtain the Mogroside-V–Melphalan ester conjugate, wherein the method (100) is performed as a one- pot, solvent-free process without an intermediate purification step.
