Abstract: The present invention relates to a recombinant protein production and purification system (100) for expression and purification of tagless recombinant human interleukin-6 (rhIL-6) using a sodium chloride inducible bacterial expression platform. The system (100) comprises an Escherichia coli strain GJ1158 host configured for osmotic stress-responsive expression of chromosomally encoded T7 RNA polymerase, a recombinant pRSET-B vector carrying a human interleukin-6 gene under control of a T7 promoter, an induction stage configured for sodium chloride mediated recombinant protein expression, and a purification stage employing affinity chromatography followed by size exclusion chromatography. The system (100) enables intracellular production of tagless recombinant human interleukin-6 without affinity fusion tags or post-expression cleavage procedures. Further, the purified recombinant protein is characterized using SDS-PAGE, western blotting, ELISA, and MALDI-TOF analysis for validation of molecular identity and purity.Fig1
