Abstract: METHOD FOR DETECTION OF RABEPRAZOLE SODIUM AND DOMPERIDONE IN COMBINED DOSAGE FORM The present invention relates to a method for the detection and quantification of Rabeprazole Sodium and Domperidone in combined dosage forms using high-performance liquid chromatography (HPLC). The method employs a mobile phase composed of acetonitrile and phosphate buffer (pH 7.4) in a 35:65 ratio, passed through a 0.45 µm filter and degassed. The chromatographic separation is achieved using a stainless-steel column (25 cm × 4.6 mm) packed with octadecylsilane bonded to porous silica (5 µm). The detection is performed at a wavelength of 280 nm with a flow rate of 1.0 ml/min and an injection volume of 20 µl. The standard solution is prepared by dissolving known quantities of Rabeprazole Sodium and Domperidone working standards in methanol and diluting with the mobile phase. Similarly, the test solution is prepared by dissolving powdered pellets of the dosage form in methanol, sonicating, filtering, and diluting with the mobile phase. The method provides accurate, reliable, and reproducible results for the quantification of active pharmaceutical ingredients (APIs) in the dosage form. This invention is particularly beneficial for quality control and assurance in pharmaceutical formulations containing these compounds.
