Abstract: ABSTRACT Disclosed herein is a method (100) for detecting trace bioethanol in a crude fermentation sample that comprises acquiring a crude fermentation broth comprising ethanol generated from fermentation of biomass, concentrating the crude fermentation broth to obtain a distilled/partially purified broth, preparing a reaction mixture by combining a predetermined volume of the distilled broth with an acidified potassium dichromate solution, maintaining the potassium dichromate solution at a concentration effective for detecting trace ethanol levels, incubating the reaction mixture under controlled temperature conditions to permit oxidation of ethanol in the presence of acidic medium, allowing reduction of dichromate ions to chromium ions causing a detectable colour transition from orange to green or bluish green, measuring the colour change by spectrophotometric analysis at a predetermined wavelength and determining presence and/or concentration of bioethanol based on absorbance intensity relative to a calibration standard.
