Abstract: This invention pertains to the marine algal bioactive compounds that can be used to manage metabolic disorders by regulating glucose metabolism, lipid metabolism, insulin signaling, oxidative stress, and metabolic inflammation. Marine algae offer an abundance of bioactive compounds having a wide variety of structures, including polyphenols, phlorotannins, carotenoids, terpenoids, sterols, fatty acids, peptides, polysaccharides, and sulfated polysaccharides, some of which have shown promise against diabetes, obesity, dyslipidemia, and metabolic syndrome. This invention provides for a systematic methodology for discovering marine algal compounds that have the potential for targeting the metabolic diseases of interest. The methodology includes preparing or selecting a library of marine algal compounds, identifying therapeutic protein targets that have relevance to the disease states, performing structure and binding site analysis of the selected targets, and evaluating the algal compounds through the process of structure-based virtual screening and molecular interaction analysis. Candidate compounds with promising target interactions can be characterized using the following methods: molecular dynamics simulation, binding energy calculation, physicochemical characterization, pharmacokinetic prediction, drug-likeness prediction, and toxicity prediction. After selection, the chosen compounds can be further validated using biochemical, cellular, or biological assays to assess their efficacy in the modulation of glucose transport, carbohydrate digestion, insulin signaling, lipid storage, cholesterol metabolism, oxidative stress, and/or other metabolic processes. The present invention includes compounds isolated from green, red, and brown marine algae, which include individual compounds, fractions, extracts, derivatives, and combinations thereof. The compounds can be used in the development of pharmaceuticals, nutraceuticals, functional foods, and therapeutics for the treatment of type 2 diabetes, obesity, dyslipidemia, metabolic syndrome, insulin resistance, nonalcoholic fatty liver disease, and other metabolic conditions.
