Abstract: The present invention discloses an isopimpinellin-based antibacterial composition for destabilizing the outer membrane of Salmonella enterica serovar Typhi. The composition comprises isopimpinellin and a microbiologically acceptable carrier configured to disperse, solubilize, or present isopimpinellin for effective contact with the bacterial outer membrane. The composition is configured to interact with lipopolysaccharide-rich regions, phospholipid domains, and outer membrane protein-associated portions of the bacterial envelope, thereby increasing membrane permeability, reducing membrane integrity, and inhibiting bacterial viability under controlled in vitro conditions. The invention further provides an in vitro evaluation arrangement comprising a bacterial culture preparation unit, a treatment unit, and an analysis unit for determining outer membrane disruption parameters including dye uptake, membrane potential change, leakage of cellular contents, viable count reduction, and membrane morphology alteration. The invention provides a membrane-directed antibacterial platform useful for evaluating the inhibitory potential of isopimpinellin against the outer membrane of Salmonella enterica serovar Typhi.
