Abstract: ABSTRACT The present disclosure relates to the isolation and characterization of endophytic fungi from leaves of Bacopa monnieri Linn., preparation of fungal extracts, and evaluation of their in vitro antioxidant activity. The leaves are surface-sterilized using 90% alcohol and embedded in a sterilized, solidified culture medium. The inoculated medium is incubated for seven days to promote endophytic fungal growth, followed by subculturing to obtain pure fungal isolates. The isolates are subjected to polymerase chain reaction (PCR)-based amplification and sequence analysis and identified as belonging to the genus Aspergillus. The fungal isolates are subsequently cultured by fermentation, and the resulting fungal broth is extracted using ethyl acetate and n-butanol to obtain respective solvent extracts. The extracts are dried, their percentage yields are determined, and preliminary phytochemical screening is performed to identify secondary metabolite classes. The screening reveals the presence of alkaloids, glycosides, steroids, flavonoids and tannins. Antioxidant activity of the ethyl acetate and n-butanol extracts is evaluated using the 2,2- diphenyl-1-picrylhydrazyl (DPPH) radical-scavenging assay, hydroxyl radical-scavenging assay and reducing power assay. The ethyl acetate extract exhibits greater antioxidant activity than the n-butanol extract under the tested in vitro conditions. The disclosed process provides a method for obtaining antioxidant-active extracts from endophytic fungi associated with Bacopa monnieri leaves. The findings support further isolation and characterization of the constituents responsible for the observed activity, including investigation of the contribution of phenolic compounds.
