Abstract: ABSTRACT OF THE INVENTION: The present invention relates to a novel ion, ically stabilized composite nanofibric patch and an environmentally friendly fabrication method designed for site-specific and pH-responsive cancer therapy. The patch comprises an interpenetrating polymer network of Polyvinyl Alcohol (PVA) and Sodium Alginate (SA), incorporating in-situ synthesized silver nanoparticles (AgNPs) and the chemotherapeutic drug Capecitabine. The silver nanoparticles are generated directly within the sodium alginate matrix using the native hydroxyl and carboxyl functional groups of alginate under controlled thermal or ultraviolet activation, thereby eliminating the need for toxic chemical reducing agents and minimizing nanoparticle aggregation. The resulting therapeutic formulation is electrospun into a nanofiber mat and subsequently stabilized through a non-destructive alcohol-phase ionic cross-linking process. The cross-linking is achieved using a 3% w/v calcium chloride (CaCl2) solution prepared in an ethanol-water co-solvent system (70:30 v/v), which enables efficient ionic stabilization while preserving the porous nanofiber morphology. The stabilized patch retains an average fiber diameter of approximately 150-300 nm and exhibits excellent structural integrity. The ionically cross-linked nanofibric patch demonstrates pH-responsive swelling and controlled drug-release behavior, releasing Capecitabine at an accelerated rate in the acidic tumor microenvironment (pH 5.5-6.5) while maintaining slower release under normal physiological conditions (pH 7.4). The combined action of Capecitabine and silver nanoparticles provides enhanced anticancer efficacy through a synergistic therapeutic mechanism, making the invention particularly suitable for localized and targeted oncological treatment.
