Abstract: The present invention relates to a chromatography column system (100) comprising a column tube (102), a movable adaptor assembly (106), an adaptor positioning mechanism (110) having a threaded drive shaft (114) and a replaceable, non-rotating dry-running load-transfer element (118), one or more process valves (174, 176), and an adaptor positioning controller (194). The controller (194) coordinates movement of the movable adaptor assembly (106) with operation of the process valves (174, 176), whereby movement of the adaptor assembly (106) varies the internal volume of the column tube (102) to generate a pressure differential for drawing process liquid into the column while permitting displacement of gas therefrom, thereby facilitating self-priming without requiring a dedicated external priming pump. The system may further comprise synchronized drive assemblies (150, 152) and position and pressure sensing arrangements for controlled adaptor positioning and fluid management. FIG. 1.
