Abstract: The present disclosure provides an in-vitro spectrophotometric method for quantitative estimation of cyanide in whole blood, wherein a preserved blood sample is acidified to liberate cyanide, which diffuses into an alkaline medium and is sequentially reacted with chloramine-T, pyridine, and barbituric acid to form a coloured complex measurable at 531 nm for quantification against calibration standards. The method employs potassium tetracyanozincate as a safer cyanide reference compound that releases cyanide only under acidic conditions, thereby improving safety, stability, and analytical reliability compared to conventional methods. The disclosure further includes a corresponding calibration reagent composition and a kit comprising requisite reagents and a microdiffusion apparatus for accurate and reproducible cyanide analysis in whole blood matrices
