Abstract: The present invention discloses a biomimetic nanovesicle, mMOF-PSL, for the treatment of triple-negative breast cancer. The nanovesicle comprises an iron-based MIL-101(Fe) metal-organic framework core co-loaded with a hypoxia-cleavable IMCA-6-azobenzene-squalene prodrug and the NIR-II photosensitizer BBTD-Ce6, encapsulated within a pH-sensitive DSPE-PEOz2K phospholipid bilayer and camouflaged with a homologous cancer-cell membrane. NIR-II irradiation at 1064 nm activates BBTD-Ce6 to consume oxygen, aggravate hypoxia and reductively cleave the azobenzene linker, releasing IMCA-6 that depletes GPX4. Concurrent Fenton catalysis by the MOF iron centres amplifies lipid peroxidation, driving ferroptosis and immunogenic cell death. The four components act co-operatively rather than additively, with a Chou-Talalay combination index of 0.07, achieving over 88 percent tumour inhibition with minimal systemic toxicity.
