Abstract: The healing of the wound is a multiphase, multifactorial physiological process that involves a balanced microenvironment, infection control, and long-term therapeutic effect to ensure optimal healing. In the current study, it is proposed to design a baicalein-loaded hydrocolloid patch (BAIHCP) incorporating a sustained release polymeric matrix to create a multifunctional phytochemical-based wound care system. The therapeutic agent (baicalein) was chosen as a bioactive flavonoid because it has strong anti-inflammatory, antioxidant and antimicrobial effects, which stimulate tissue repair and alleviate oxidative stress of the wound site. Biocompatible polymers that could retain moisture, create a protective occlusive barrier, and release the drugs over a long period were used to design the hydrocolloid patch. The approach to formulation was to entrap baicalein within a controlled release polymer network to provide a slow, continuous delivery over a long time, and thus, reduce the dosing time and increase the therapeutic effect. The structural integrity and functionality of the patch was validated by physicochemical characterization, SEM, FTIR, XRD, Contact angle, and TGA. The in vitro-like conditions of drugrelease at various temperature and pH conditions were simulated and supported a sustainedrelease profile to meet the wound healing needs. Biological assessment revealed that the BAIHCP had great antimicrobial. The developed patch is an integrated wound care system that involves the benefits of hydrocolloid technology and therapeutic efficacy of baicalein. This versatile solution provides a viable alternative to traditional wound dressings by enhancing healing, minimising the risk of infection, and enhancing comfort to the patient with fewer dressing changes and regulated phytochemical release.
