Abstract: The present invention relates to a pharmaceutical co-crystal preparation and characterization system (100) for producing Gemcitabine hydrochloride co-crystals exhibiting enhanced physicochemical properties and improved anticancer performance. The system (100) comprises an active pharmaceutical ingredient storage unit (102), a co-former storage unit (104), a weighing and proportioning module (106), a liquid-assisted grinding assembly (108), a humidity-controlled processing chamber (110), an in-line particle-size verification module (112), a characterization unit (114), and a processing controller (116). Gemcitabine hydrochloride is co-crystallized with triethanolamine, adipic acid, or cinnamic acid through a liquid-assisted grinding process performed under controlled environmental conditions. The prepared co- crystals are characterized using Fourier-transform infrared spectroscopy, differential scanning calorimetry, pH analysis, dissolution studies, solubility studies, assay measurements, and in-vitro anticancer evaluation. The humidity-controlled chamber (110) and the particle-size verification module (112) improve crystal reproducibility and formulation consistency. The invention provides enhanced solubility, dissolution behavior, stability, and pharmaceutical suitability without chemically modifying the active pharmaceutical ingredient
