Abstract: Embodiments herein relates to a hemostatic sponge, a composition, and a method for preparation thereof. The sponge according to embodiments herein includes a porous, cross-linked matrix, wherein the matrix comprises a protein–polysaccharide network having covalent amide linkages forming a three-dimensional interconnected structure. Embodiments herein further disclose a composition for preparing the hemostatic sponge. In one embodiment, the composition includes a protein; a polysaccharide; a coagulation cascade activator; and a cross-linking system. The disclosed sponge exhibits a porous and absorbent structure capable of rapid fluid uptake and effective interaction with blood components to promote rapid clot formation. Further, the disclosed sponge demonstrates improved blood clotting performance along with favorable swelling behavior and compatibility with blood cells. Furthermore, the disclosed sponge is cost-effective, scalable, and suitable for use in surgical, trauma, and emergency care applications. FIG. 1
