Abstract: ABSTRACT The present disclosure relates to handheld apparatus and methods for rapid DNA profiling from trace biological samples. The handheld apparatus (102) includes a magnet actuator (104) operatively coupled to a microfluidic cartridge (106), a thermoelectric circuit (108) thermally coupled to a polymerase chain reaction reactor (110) within the microfluidic cartridge (106), an optical image sensor (112) positioned adjacent to the polymerase chain reaction reactor (110), and a hardware processor (114). The hardware processor (114) signals the magnet actuator (104) to elute extracted DNA into the polymerase chain reaction reactor (110) and signals the thermoelectric circuit (108) to thermally cycle the reactor (110), amplifying short tandem repeat loci. The optical image sensor (112) captures melt-curve fluorescence data across spatially distinct locus zones. The hardware processor (114) computes a short tandem repeat profile by identifying allele-characteristic inflection points against corresponding melting temperature values and transmits the profile to a display screen (116).
