Abstract: The present invention relates to a Gemcitabine hydrochloride-loaded graphene quantum dot-doped metal-organic framework-5 (GQD@MOF-5) nanocomposite for breast cancer therapy. Graphene quantum dots are prepared by hydrothermal pyrolysis, conjugated with MOF-5, and subsequently loaded with Gemcitabine hydrochloride. The formulation is optimized using a Box-Behnken design by varying GQD, MOF-5, and drug amounts and evaluating entrapment efficiency and drug loading. The developed nanocomposite is characterized by DSC, FTIR, XRD, SEM, EDX, and related physicochemical techniques. In vitro release studies demonstrate sustained and pH-dependent Gemcitabine release under physiological and acidic conditions. Cytotoxicity studies against MCF-7 breast cancer cells show an IC50 of 10.47 µg/mL for the drug-loaded formulation compared with 26.9 µg/mL for free Gemcitabine. Fluorescence microscopy demonstrates enhanced cellular uptake, while apoptosis studies reveal characteristic nuclear fragmentation and chromatin condensation. The nanocomposite thereby provides a multifunctional platform for controlled Gemcitabine delivery with enhanced in vitro anticancer activity.
