Abstract: Gold nanoparticles have emerged as high-efficient nanocarriers for advanced drug-delivery systems for their unique physicochemical properties including high surface area and biocompatibility. This invention focuses on, synthesis, characterization, and evaluation of gold nanoparticle-based drug-delivery systems for improved therapeutic performance. Gold nanoparticles were successfully synthesized by citrate reduction method, indicated by a characteristic surface plasmon resonance peak at approximately 525nm and a visible color change to ruby red. The nanoparticles exhibited uniform spherical shape with average size range of 25-45 nanometers and good colloidal stability as confirmed by zeta potential values of negative 22 to negative 28 millivolts. In vitro release analysis showed controlled and sustained drug release at physiological pH 7.4, while significantly faster release was observed under acidic conditions pH 5.5, indicating suitability for tumor-targeted drug delivery. Cytotoxicity results suggested enhanced anticancer activity of drug-loaded gold nanoparticles compared to free drug, while bare gold-nanoparticles exhibited minimal toxicity, confirming biocompatibility.
