Abstract: The present invention discloses a glycan-functionalized bacterial outer membrane vesicle nanosystem designated HA-Man-OMVs@DOX/SOR for synergistic chemo- immunotherapy of hepatocellular carcinoma. The nanosystem comprises outer membrane vesicles isolated from Escherichia coli, surface-engineered with DSPE- PEG2000-hyaluronic acid and DSPE-PEG2000-mannose conjugates through lipid-anchored post-insertion. Hyaluronic acid targets CD44-overexpressing tumor cells and cancer stem cells while mannose targets CD206 on tumor-associated macrophages to reprogram immunosuppressive M2 phenotype to anti-tumor M1 phenotype. Doxorubicin is encapsulated in the aqueous lumen and sorafenib is intercalated in the lipid bilayer achieving dual-compartment co-delivery with synergistic anti-tumor efficacy demonstrated by a combination index of less than one. The nanosystem exhibits pH-responsive drug release, enhanced cellular uptake, significant tumor growth inhibition of 84.6 percent in xenograft models, and favourable biosafety. The invention also discloses the process for preparing the said nanosystem.
