Abstract: ABSTRACT GENETICALLY EXPANDED T7-RNA-POLYMERASE (SSBT7-RNAP): FLUORESCENT TRIAZOLYL CYANOBENZENE ALANINE (TCNBALAAC) UNNATURAL AMINO ACID AT Y571 CODON SITE The invention relates to the site-specific incorporation of a novel fluorescent triazolyl unnatural amino acid (FTUAA) into T7 RNA polymerase using genetic code expansion (GCE) techniques, more particularly, it address one critical site is the Y571 position, a mutation hotspot adjacent to the enzyme’s active site, where substitutions such as Y571S (tyrosine to serine) typically lead to compromised activity and lower transcriptional efficiency.
