MUMBAI, India, Aug. 24 -- Intellectual Property India has published a patent application (202641097383 A) filed by Vellore Institute Of Technology on August 12, 2026, for Genetically Engineered Mammalian Cells And Methods For Targeted Transgene Integration At Vwf Exon 52.
Inventors include Balaji Balakrishnan; and Nishal Kumarasamy.
The application for the patent was published on August 21, 2026, under issue no. 34/2026.
Abstract: A genetically engineered mammalian cell (112) and a method (700) for generating the genetically engineered mammalian cell (112) are disclosed. The genetically engineered mammalian cell (112) comprises a mammalian host cell (102) having a recombinant transgene integrated at an exon 52 locus (108) of a von Willebrand factor (VWF) gene by a site-directed genome editing system (104). The genome editing system (104) creates a site-specific double-strand break at the exon 52 locus (108), followed by integration of donor DNA (106) comprising a recombinant transgene encoding a reporter protein, a therapeutic protein, or a recombinant protein. In some embodiments, the mammalian host cell (102) comprises a HEK293T cell, and the recombinant transgene (106) comprises a ZsGreen1 reporter gene expression cassette or a B-domain deleted factor VIII (BDD-FVIII) expression cassette operably linked to a CAG promoter. Selected cells are expanded to obtain stable mammalian producer cells for sustained recombinant protein production. FIG. 1
