Abstract: Gastroretentive Floating Tablets for Sustained Pantoprazole Sodium Release to Treat Gastroesophageal Reflux Disease This invention describes the development and optimization of gastroretentive floating tablets for the sustained release of pantoprazole sodium, a proton pump inhibitor widely used in the treatment of gastroesophageal reflux disease (GERD) and related gastric conditions. The tablets are designed to achieve prolonged gastric retention and controlled drug release through the integration of hydrophilic polymers, such as hydroxypropyl methylcellulose (HPMC), and gas-generating agents, including sodium bicarbonate and citric acid. Upon contact with gastric fluids, the gas-generating agents produce carbon dioxide, which becomes entrapped within the gel matrix formed by HPMC, ensuring buoyancy and sustained release. The formulation is optimized to exhibit a floating lag time of less than 30 seconds, buoyancy exceeding 10 hours, and a drug release profile achieving 85.09% release within 6 hours. This system offers enhanced bioavailability, reduced dosing frequency, and improved therapeutic outcomes, providing a patient-compliant and scalable solution for advanced drug delivery.
