Abstract: ABSTRACT “Gallic Acid Loaded Microsponge Formulation for Sustained Topical Anti-Inflammatory Therapy and Process for Preparation Thereof” The present invention discloses a pharmaceutical composition comprising gallic acid-loaded microsponges for sustained topical anti-inflammatory therapy. The microsponges are prepared by a quasi-emulsion solvent diffusion technique employing Eudragit LS-100 and ethyl cellulose as polymeric carriers. The gallic acid is entrapped within the porous polymeric microsponge matrix, which controls and sustains drug release upon topical application. The microsponges exhibit porous near- spherical morphology with pore sizes of 1.4–4.5 µm, particle sizes of 5–200 µm, entrapment efficiency of approximately 60%, and a percentage yield of 60%. The microsponges are incorporated into an oil-in-water cream base to yield a formulation with skin- compatible pH (6.0–6.5), appropriate viscosity (59,160 cP), good spreadability, and cosmetic elegance. In-vitro studies demonstrate sustained release of approximately 98% gallic acid over 8 hours following first-order kinetics. The formulation exhibits 48.92% inhibition of protein denaturation, confirming effective anti-inflammatory activity. The invention provides improved drug stability, enhanced skin retention, reduced dosing frequency, minimized irritation, and improved patient compliance compared to conventional topical anti-inflammatory formulations.
