Abstract: Formulation of 3D nanofibrous scaffold dressing material for diabetic burn wound and the process thereof The present invention relates to a poly(vinyl alcohol) (PVA) and chitosan-based three-dimensional (3D) bioactive nanofibrous scaffold developed as an advanced wound dressing material for diabetic burn wound healing. The scaffold comprises PVA in a concentration range of 7–10% w/v, chitosan in a concentration range of 1–3% w/v, Rosmarinic acid (RA) in a concentration range of 0.5– 1.5 mg/mL, and epidermal growth factor (EGF) in a concentration range of 10–100 µg/mL. The scaffold is fabricated through electrospinning followed by post- fabrication transformation involving fragmentation, homogenization, sequential freezing, lyophilization, and thermal cross-linking to obtain a porous 3D interconnected architecture. The developed scaffold exhibits fiber diameter ranging from 80–150 nm, porosity of 50–60%, controlled water vapor transmission rate, high swelling capacity, enhanced drug loading efficiency, antibacterial activity, antioxidant activity, biocompatibility, non-haemolytic behaviour, and controlled biodegradation. The synergistic incorporation of RA and EGF promotes cellular proliferation, angiogenesis, and accelerated tissue regeneration, thereby making the scaffold highly suitable for diabetic burn wound management.
