Abstract: The present invention relates to a polyherbal bilosomal formulation comprising silymarin and bitter melon (Momordica charantia) extract for the management of metabolic dysfunction-associated steatotic liver disease (MASLD). The bilosomal formulation comprises silymarin, bitter melon extract, soya phosphatidylcholine, cholesterol, and sodium deoxycholate as a bile salt. The bilosomes are prepared by a thin-film hydration technique, wherein silymarin is incorporated into the lipid phase and bitter melon extract is incorporated through the aqueous hydration phase. The formulation is optimized using a Box–Behnken statistical design considering phosphatidylcholine, cholesterol, and sodium deoxycholate as formulation variables and vesicle size and entrapment efficiency as responses. The resulting bilosomal system provides effective entrapment and controlled release of the incorporated phytoconstituents. The combined delivery of silymarin and bitter melon provides hepatoprotective, anti-steatotic, antioxidant, anti-inflammatory, and metabolic regulatory effects, thereby providing a polyherbal nanovesicular approach for the management of MASLD and associated hepatic metabolic dysfunction.
