Abstract: The present invention relates to a herbal niosomal intranasal spray formulation comprising Valeriana officinalis extract and Humulus lupulus extract incorporated within niosomal vesicles comprising a non-ionic surfactant and cholesterol. The niosomal vesicles are prepared by a thin-film hydration technique followed by sonication and separation, and are incorporated into an aqueous nasal spray vehicle comprising chitosan, glycerin, sodium chloride, benzalkonium chloride and purified water. In one embodiment, the formulation comprises approximately 200 mg each of Valeriana officinalis and Humulus lupulus extracts per 200 mL. The formulation exhibits an entrapment efficiency of approximately 91.88%, a Z-average particle size of approximately 198.4 nm and a polydispersity index of approximately 0.312. The formulation further provides suitable physicochemical characteristics, substantially uniform spray characteristics and progressive release of the incorporated herbal constituents. The invention provides a convenient herbal vesicular dosage form for intranasal administration and is contemplated for applications associated with calming, anxiolytic and sleep-related effects, with potential for delivery through nasal pathways towards the central nervous system.
