Abstract: ABSTRACT [0024] Miconazole, an imidazole antifungal agent, has been widely recognized for its broad-spectrum activity against fungal pathogens, including Candida species. It exerts its antifungal effect by inhibiting the biosynthesis of ergosterol, a crucial component of fungal cell membranes, thereby disrupting membrane integrity and leading to cell death. Miconazole’s efficacy and safety profile make it a preferred choice in treating various superficial fungal infections, including oral candidiasis. However, issues such as limited bioavailability and patient compliance necessitate the development of more effective and patient-friendly formulations. Lozenges represent a suitable dosage form for delivering antifungal agents locally within the oral cavity. They offer several advantages, including prolonged contact time with the mucosal surfaces, ease of administration, and improved patient compliance, especially among paediatric and geriatric populations. The controlled release of miconazole through lozenges can potentially enhance its therapeutic efficacy by maintaining adequate drug concentration at the site of infection for extended periods. Furthermore, lozenges can provide a practical solution to overcome the quick clearance typically experienced with oral rinses or gels. [0025] The formulation of miconazole lozenges requires careful consideration of factors such as drug stability, release characteristics, palatability, and patient acceptability. Evaluation of these lozenges is equally important to ensure their quality, safety, and therapeutic effectiveness. Parameters like dissolution rate, antifungal activity, mechanical properties, and in vitro performance need to be rigorously assessed. This project aims to develop and evaluate miconazole lozenges tailored for the treatment of oral thrush. The study focuses on optimizing the formulation to enhance drug release and retention in the oral cavity, ensuring effective antifungal action with minimal side effects. By addressing the formulation challenges and conducting comprehensive evaluations, this research strives to contribute toward improved management of oral candidiasis, enhancing patient outcomes and treatment experiences.
