Abstract: 3. Abstract: FORMULATION AND EVALUATION OF LEMONGRASS (Cymbopogon citratus) EXTRACT-LOADED SODIUM ALGINATE NANOPARTICLE MICRONEEDLE PATCH FOR LUMBAR DISC BULGE PAIN MANAGEMENT. The present invention relates to the formulation and evaluation of a biodegradable transdermal microneedle patch comprising sodium alginate nanoparticles loaded with lemongrass extract derived from Cymbopogon citratus for the management of lumbar disc bulge pain and inflammation. The invention combines herbal therapeutics, nanotechnology, and microneedleassisted transdermal drug delivery to provide localized, sustained, and minimally invasive pain management. The bioactive constituents oflemongrass, including citra!, flavonoids, terpenoids, and phenolic compounds, possess significant anti-inflammatory, analgesic, antioxidant, and muscle relaxant activities. However, conventional topical formulations exhibit limited skin penetration and poor bioavailability.in the present invention, lemongrass extract is encapsulated within sodium alginate nanoparticles prepared by ionic gelation using calcium chloride as a crosslinking agent. The nanoparticles are characterized for particle size, encapsulation efficiency, zeta potential, and drug release behavior. The prepared nanoparticles are incorporated into a dissolvable microneedle matrix fabricated using biodegradable polymers such as polyvinyl alcohol, polyvinylpyrrolidone, sodium alginate, gelatin, and hydroxypropyl methylcellulose. Upon application over the lumbar region, the microneedles penetrate the stratum corneum and dissolve gradually, enabling controlled and sustained release of phytoconstituents into localized tissues.the developed formulation demonstrates enhanced transdermal permeation, prolonged therapeutic activity, improved patient compliance, and reduced systemic side effects compared with conventional oral or topical therapies. The invention provides a safe, effective, and patient-fi·iendly approach for localized treatment of lumbar disc bulge-associated lower back pain and inflammation.
