Abstract: The present invention relates to a topical ketoprofen loaded emulgel composition for enhanced skin permeation and anti-inflammatory activity. The composition comprises ketoprofen at 2% as active drug, Carbopol 934 at 0.5% to 2% as gelling agent, liquid paraffin at 7.5% as oil phase, Span 80 and Tween 80 each at 2% as surfactant pair, propylene glycol at 3% to 7% as penetration enhancer, methyl paraben at 0.2% as preservative, triethanolamine for pH adjustment and purified water as vehicle. The optimised emulgel exhibits skin-compatible pH of 6.4, viscosity of 23,400 cP, spreadability of 24.3 g·cm/s, drug content of 99.1%, in-vitro drug release of 98% in 8 hours, ex-vivo skin permeation of 74%, and approximately 75% inhibition of carrageenan-induced paw edema at 6 hours, providing superior anti-inflammatory activity over standard gels.
